Aging is associated with the accumulation of cellular toxins and damage. Declines in cellular detoxification mechanisms and impairments in antioxidant protection are consistently
observed in aging models and likely contribute to ageassociated accumulation of cellular damage. The master regulator, Nuclear factor erythroid 2-related factor 2 (Nrf2), is a transcription factor that regulates the basal and inducible expression of a large battery of genes encoding for
cytoprotective factors including those that defend against electrophilic stressors and oxidative insults. The Nrf2/electrophile response element (EpRE) Antioxidant Protection and Phase II Detoxification pathways are impaired with aging due to age-related changes in gene expression (2). A key example is the reduction in glutathione (GSH) levels in all tissues with age due primarily to declines in glutamatecysteine ligase and glutathione synthase expression (1). Opposing these changes in gene expression may delay or attenuate the aging process. Most anti-aging intervention strategies to date, have tested single ingredients and have focused solely on an individual gene, in an isolated tissue. A more comprehensive strategy is to examine changes in the expression patterns of multiple genes or pathways in specific tissues and then to identify a
blend of phytochemicals that opposes those age-related changes. It seems prudent to examine changes that occur during middle age rather than waiting until old age when an intervention may not be as impactful. Accordingly, the purpose of this study was to test a blend of natural
compounds in middle-aged mice, compared to young mice, for the ability to oppose age-related changes in the expression of Nrf2-regulated genes involved in the detoxification of xenobiotics and xenobiotic metabolites and in the synthesis and regulation of intrinsic antioxidants and
antioxidant enzymes.

Abstract.

Aging is characterized by a gradual loss in physical and mental function starting at 30-40 years of age. Thepurpose of this study was to compare a novel combination of Cordyceps sinensis, pomegranate extract, andPanax ginseng (TRT) to control (CNTL) formula regarding physical, mental, and sexual desire. Healthy malesand female subjects (n=116) were recruited to participate in a prospective, double-blind, randomized,placebo-controlled trial that lasted 60 days. Subjects were randomized into a control (n= 56; CNTL) ortreatment (n= 60; TRT) groups and stratified by age and gender. Mental (MCS) and physical (PCS) wellbeing were measured by validated questionnaires. Other measurements included blood chemistry measurements,
skin carotenoid concentrations, and adverse events. There were n=48 in CNTL and n=47 in TRT groups that completed the study. No differences were detected in the mean scores for (PCS=CNTL;-0.47, TRT;-0.08) or (MCS=CNTL;0.32, TRT;-1.98) and sexual desire between groups nor between age. There was a strong trend towards higher skin carotenoid concentrations in the TRT group (P=0.052). There was no difference between groups for adverse events or blood chemistry. The TRT formulation appears to be safe. We were unable to detect differences in physical, mental, and sexual desire as seen in animal studies presumably because this
population was so healthy and didn’t show an age related decline. There was an interesting finding that subjects consuming the TRT formulation experienced a trend towards higher skin carotenoid concentrations and warrants further investigation in a population which has age-related decline in physical, mental, sexual decline, or experiencing greater oxidative stress.

Summary
We used DNA microarrays to identify panels of transcriptional markers of aging that are differentially expressed in young (5 month) and old (25 month) mice of multiple inbred strains (129sv, BALB ⁄ c, CBA, DBA, B6, C3H and B6C3F1). In the heart, age-related changes of five genes were studied throughout the mouse lifespan: complement component 4, chemokine ligand 14, component of Sp100-rs, phenylalanine hydroxylase and src family associated phosphoprotein 2. A similar analysis in the brain (cerebellum) involved complement component 1q (alpha polypeptide), complement component 4, P lysozyme structural, glial fibrillary acidic protein and cathepsin S.Caloric restriction (CR) inhibited age-related expression of these genes in both tissues. Parametric analysis of gene set enrichment identified several biological processes that are induced with aging in multiple mouse strains. We also tested the ability of dietary antioxidants to oppose these transcriptional markers of aging. Lycopene, resveratrol, acetyl-L-carnitine and tempol were as effective as CR in the heart, and a-lipoic acid and coenzyme Q10 were as
effective as CR in the cerebellum. These findings suggest that transcriptional biomarkers of aging in mice can be used to estimate the efficacy of aging interventions on a tissue-specific basis. Key words: aging; antioxidants; biomarkers; caloricrestriction; microarray; mouse.
